Effect of sensitizing temperature and time on helper T lymphocytes quantitation


Abstract
Peripheal blood from 34 normal healthy Korean subjects was examined for the quantita= tion of helper T lymphocytes. We counted the helper T lymphocytes by using the conventional double rosette mothod and modified sensitizing method. It is supposed that the factor
affecting the number of helper T lymphocytes is sensitizing temperature and time.
The procedures for the sensitizing temperature and time during the sensitization of ox RBC with anti ox RBC rabbit Ig M in the conventional double rosette method was done for 2 hours at room temperature, while in the modified method, for 30 mintes at 37¡ÆC water bath. We compared statistically the resulting data by using the conventional and modified sensitizing method. The results are as follows.
1. Means of helper T lymphocytes were 28.6¡¾7.1% in the coventional double rosette mothod.
2. Means of helper T lymphocytes were 42.2¡¾8.000 in the modified sensitizing method. 3. This difference is statistically significant (p<0.005).
